The sample mix undergoes droplet formation so that the DNA strands are compartmentalized into separate droplets. Then, they are amplified via PCR, and the amplified product reacts with the fluorescence-labeled probe. Each droplet is scanned and plotted in its corresponding coordinates. The x-axis represents signal detected from Channel 2 (VIC/HEX), while the y-axis represents signal detected from Channel 1 (FAM)
